Bradford surpassed the previous high of 429 yards, set by Josh Heupel on September 25, 1999 at Louisville.
Juaquin Iglesias had 12 receptions for 191 yards for the Sooners (6-1, 2-1 Big 12), who rebounded from a 45-35 setback to perennial rival Texas last week, which knocked the team from the No. 1 ranking.
Quentin Chaney picked up 105 yards on six catches, while DeMarco Murray racked up 83 yards and two scores on 16 carries. Chris Brown added 92 yards and another touchdown on the ground.
"Bradford was sensational, Juaquin Iglesias had a huge game and both backs really ran the ball well," said Oklahoma head coach Bob Stoops. "It was really solid. Defensively, there were parts when we really gelled in the second half. These guys have put a ton of points and yards on everybody."
Todd Reesing completed 24-of-41 passes for 342 yards, two touchdowns and a pair of interceptions for the Jayhawks (5-2, 2-1), who suffered their first defeat in conference play since a memorable loss at the hands of Missouri last November when Kansas was the second-ranked school in the nation.
Dezmon Briscoe had a monster game with 12 catches for 269 yards receiving, including three scores. Jake Sharp added 103 yards on the ground, with a TD, on 12 carries.
After the Red River Shootout between Texas and Oklahoma featured 873 yards of total offense, Kansas and Oklahoma combined to amass 1,165 yards on Saturday.
"The O-line was getting pushed," said Brown. "They were getting up the field and making big, gaping holes. Whenever they didn't make them, we still tried to do what we could."
Kansas failed on its first drive of the second half, and Oklahoma countered by going up 31-17 on Bradford's six-yard connection to Jermaine Gresham. The drive covered 53 yards in only three plays.
Quick strikes continued as Kansas took less than two minutes off the clock to get within 31-24 on Sharp's 17-yard scoring run, but Murray's seven-yard rush put Oklahoma back on top by 14 with 6:14 remaining in the third.
Bradford hit Kolby Smith from the two with 12:02 to play in the contest to boost the home team to a 45-24 advantage, and the Oklahoma defense began to close in on Reesing and force him to rush throws.
Kansas was unable to respond until a Reesing 17-yard TD pass to Briscoe with under a minute to go.
"We came in expecting to have to score a lot of points to have a chance to win," said Reesing. "We had some mistakes and penalties that slowed drives, and cost us in the long run. We were not very good on third down like we have been all year, and the turnover in the red zone didn't help.
The Sooners picked up the game's first score as Bradford hit Matt Clapp for a 12-yard touchdown. The Jayhawks countered on the following series when Jocques Crawford reached the end zone on a two-yard run.
Oklahoma forged ahead by a touchdown mere seconds into the second quarter as Murray ran in from the three to cap a 10-play, 79-yard drive.
Kansas had a golden chance to tie the game with 8 1/2 minutes left in the half, but Reesing was picked off by Lendy Holmes at the Sooners' two-yard- line. Bradford couldn't move the offense, and Kansas came right back.
A 40-yard hookup from Reesing to Briscoe put KU at the OU 16, but the Sooners held until a 23-yard field goal from Jacob Branstetter made it 14-10 with 5:16 to play in the quarter.
Brown's 14-yard high-wire act along the right sideline put Oklahoma up 21-10 with three minutes before halftime, but 25 seconds later a defensive breakdown led to Briscoe's 69-yard catch-and-run for a four-point contest.
"It always feels good that the coach is comfortable with you enough to allow you to make plays," Briscoe admitted. "I was excited when they called my number."
Jimmy Stevens' 19-yard make with four ticks left gave the Sooners a 24-17 lead at the break.
Game Notes
Oklahoma holds a 67-27-6 lead in the all-time series and has won the last five meetings...Iglesias' 10 first-half catches and 178 receiving yards both set school records. He broke the first-half yardage mark set by Mark Clayton (166) against Texas A&M 2003. The Sooners were without the services of leading receiver Manuel Johnson, who left the game early in the first quarter with an apparent arm injury and did not return...Oklahoma converted 7-of-16 third-down opportunities while holding Kansas to 2-of-12...The Sooners outrushed the Jayhawks 206-134 and picked up 36 first downs, while the visitors had 26.
