Murray carried the ball 20 times for 112 yards and caught two passes for 28 yards for the Sooners (6-0, 2-0 Big 12), who were coming off a 28-20 win over Texas on October 2.
Murray set the school's all-time touchdown mark in the process. His third touchdown of the night gave him 58 for a career, topping the mark set by 1969 Heisman Trophy winner Steve Owens.
"It's great. The Lord has blessed me tremendously," Murray said. "My offensive line has done a great job blocking and making holes, so I give credit to Jesus, and I give credit to my line."
Landry Jones connected on 30-of-34 passes for 334 yards and three touchdowns. Ryan Broyles caught 15 passes for 182 yards and a touchdown for Oklahoma, which has won 12 consecutive games against Iowa State.
Austen Arnaud completed 12-of-18 passes for 102 yards for the Cyclones (3-4, 1-2), who have lost two in a row after winning two straight.
Alexander Robinson carried the ball seven times for 39 yards, while Jake Williams caught three passes for 51 yards for Iowa State, which was outgained by a 672-183 margin.
"They're great players. They played fast, they executed well, maintained blocks," Iowa State head coach Paul Rhoads said. "You didn't see our guys getting off blocks and making plays. Much better execution by their football team."
Oklahoma started the game with a 12-play, 80-yard drive. Faced with a 3rd- and-2 from the three-yard line, Jones found James Hanna in the end zone for an early 7-0 lead.
Jimmy Stevens made a 37-yard field goal with 4:13 left in the first to make it a 10-0 game.
Iowa State got to the Oklahoma 27 early in the second period, but Grant Mahoney missed a 45-yard field goal. Broyles gave the Sooners a 17-point cushion with a 39-yard TD catch.
Trey Millard's one-yard rushing touchdown on the next Oklahoma possession made it a 24-0 contest with 4:29 left in the half. After the Cyclones failed to get a first down on 4th-and-8 from the 41-yard line of the Sooners, Murray rumbled into the end zone from one yard out for a 31-0 halftime lead.
Murray highlighted the scoring in the third quarter, catching a 15-yard TD pass and rushing for a 10-yard score to make it 45-0.
Drew Allen rushed for a seven-yard touchdown in the fourth to account for the final margin.
