Martinez was 23-of-35 for a school freshman-record 323 yards and totaled 112 yards rushing for the Cornhuskers (6-1, 2-1 Big 12), who were coming off a 20-13 home loss to Texas last weekend in a rematch of last season's conference championship game. Brandon Kinnie caught three passes, all for a touchdown.
Nebraska, which has won seven straight on the road, recorded its first triumph in Stillwater since 1995.
"I like playing road games," Martinez said. "I like the atmosphere and the hostility. I think our team comes together as a unit more when we play on the road."
Brandon Weeden completed 18-of-35 passes for 283 yards with two touchdowns, both of them going to Justin Blackmon, and one interception for the Cowboys (6-1, 2-1), who were denied an opportunity to match their second-best start in school history. Oklahoma State won its first seven games in 2008.
"Obviously it is a tough one to swallow," Weeden said. "We came out and we competed on both sides of the ball. We put our defense in some bad situations. Third downs were the difference in the game today. We didn't convert when we needed to and they did."
Kendall Hunter ran for 201 yards and a pair of scores in the loss.
The Cornhuskers converted a lengthy fourth down on their first possession en route to producing the game's first points. Facing a 4th-and-8 from his own 22-yard line, Alex Henery faked the punt and hustled 27 yards to midfield. Three plays later, Martinez and Kinnie hooked up for a 45-yard touchdown.
Dan Bailey kicked field goals of 31 and 44 yards to narrow Oklahoma State's deficit to one, but Nebraska took a 14-6 lead after Niles Paul returned the kickoff following Bailey's second field goal 100 yards for a touchdown.
The Cowboys immediately responded with an 11-play, 76-yard drive that ended with a one-yard run by Jeremy Smith late in the first quarter.
The teams combined for 31 points in the second stanza.
Nebraska led 21-13 after a one-yard TD reception by Tyler Legate, but Oklahoma State answered with an eight-yard TD run from Hunter. Henery then drilled a 52-yard field goal before Blackmon caught a Weeden pass for an 80-yard score to give Oklahoma State its first lead of the game, 27-24, with 5:59 remaining. The Cowboys, though, trailed, by four points after Kinnie's second touchdown grab -- this time from nine yards away -- came inside the final minute of the quarter.
Following the break, Henery booted a 32-yarder and Kyler Reed hauled in a 41- yard strike from Martinez to give the Cornhuskers a 41-27 lead. A one-yard TD run by Hunter whittled the deficit to seven with 3:29 to play in the third quarter.
Nebraska pulled away in the final frame as Henery converted a 45-yarder and Kinnie caught his third TD pass of the game, an eight-yard connection with 4:29 remaining.
The Cowboys got a late 25-yard TD pass from Weeden to Blackmon to account for the final score.
