Dennis Dixon completed 13-of-23 passes for 150 yards with a touchdown for the Ducks (6-2), who rebounded following a 34-23 loss at Washington State last week.
Jaison Williams caught eight passes for 112 yards and a TD for Oregon, which rolled up 431 yards of total offense.
Mu'Ammar Ali rushed 23 times for 139 yards and a score for the Vikings (5-4), while Sawyer Smith connected on 5-of-15 passes for 45 yards and a TD, but was picked off three times. Brian White went 4-of-7 passing for 48 yards.
Portland State got within 21-12 with 10:39 left in the second quarter on Ali's 57-yard TD run, but a two-point conversion try failed.
It was all Ducks from there. Johnson ran six yards for a score with 4:15 remaining until halftime and he scored from a yard out with 39 seconds left following another punt by the Vikings for a 35-12 margin at the half.
Paul Martinez booted a 48-yard field goal midway through the third quarter and A.J. Tuitele picked off Smith and returned it 26 yards for a score less than two minutes later.
Martinez added a 34-yard field goal in the fourth and Andiel Brown accounted for the final margin with a seven-yard touchdown run.
Williams caught an 11-yard TD pass from Dixon in the first and Johnson scored on a one-yard run for a 14-0 Oregon lead.
Tremayne Kirkland caught a 19-yard TD pass from Smith with 47 seconds left in the first, but the extra point try was blocked.
Brady Leaf scored on a three-yard run 2 1/2 minutes into the second for a 21-6 Oregon advantage.
Game Notes
Oregon is 4-0 at home this season and has won seven straight in Eugene dating back to last year...Oregon has won its only three previous meetings with Portland State, including a 41-0 thrashing of the Vikings back in 2002...Leaf was 4-of-9 passing for 34 yards.
